چکیده

This workshop on limb development was a highly enjoyable meeting that covered a wide range of topics related to appendage development and evolution. The discussions were vivid, open and stimulating for all participants. It was particularly interesting to see how broadly accepted concepts, such as the morphogen models in the fly and the progress zone model in vertebrates, were re-evaluated on the basis of new evidence presented. Important contributions included intense analyses of the roles of factors important in limb development, such as Wingless (Wg) in the fly and Sonic Hedgehog (Shh) in vertebrates. In addition, the overall view of appendage evolution was significantly expanded by data arising from new animal models such as dogfish and cricket and from the analysis of regulatory regions of genes involved in appendage patterning. Within the limits of this report, we have tried to condense the essentials of the advances presented at this meeting. Limb initiation and identity

Altered expression of the chicken homeobox-containing genes GHox-7 and GHox-8 in the limb buds of limbless mutant chick embryos.

It has been suggested that the reciprocal expression of the chicken homeobox-containing genes GHox-8 and GHox-7 by the apical ectodermal ridge and subjacent limb mesoderm might be involved in regulating the proximodistal outgrowth of the developing chick limb bud. In the present study the expression of GHox-7 and GHox-8 has been examined by in situ and dot blot hybridization in the developing l...

Congenic method in the chick limb buds by electroporation.

Electroporation is a powerful tool with which to study limb development. Limb development, however, remains an intricate series of events, requiring the precise dissection of developmental processes using relevant transgenes. In this review, we describe the anatomy of the limb field as the basis of targeted electroporation, and specific expression vectors are discussed. We share a useful protoc...

IGF-I, insulin and FGFs induce outgrowth of the limb buds of amelic mutant chick embryos.

IGF-I, insulin, FGF-2 and FGF-4 have been implicated in the reciprocal interactions between the apical ectodermal ridge (AER) and underlying mesoderm required for outgrowth and patterning of the developing limb. To study further the roles of these growth factors in limb outgrowth, we have examined their effects on the in vitro morphogenesis of limb buds of the amelic mutant chick embryos wingle...

Experiments on developing limb buds of the axolotl Ambystoma mexicanum.

Various experiments were performed on the limb buds of axolotls to compare the behaviour of amphibian limbs with that previously reported for chick limbs. Following removal of the tip or whole limb bud, extensive powers of regulation were observed since complete limbs always formed. Similarly after distal to proximal grafts intercalary regulation occurred to produce perfect limbs and after prox...

The Effect of Nano-Titanium Dioxide on Limb Bud Development of NMRI Mouse Embryo In Vivo

OBJECTIVE There is a wide application of titanium dioxide (TiO2) nanoparticles (NPs) in industry. These particles are used in various products, and they also has biological effects on cells and organs through direct contact. MATERIALS AND METHODS In this experimental research, the effect of TiO2 on chondrogenesis of forelimb buds of mice embryos was assessed in in vivo condition. Concentratio...
